RESEARCH INTERESTS:

My laboratories are interested in elucidating the molecular mechanism of pathogen-induced cytokine storm and lethality, and identify potential targets for immunotherapy in order to prevent tissue damage and fibrosis. We address these questions from wo aspects: Host-Pathogen Interaction and Immunomodulation.

Host-Pathogen Interaction

Initiation of inflammatory reaction is via the activation of innate immunity receptors on NK cells, monocytes, macrophages, PMNs and platelets by either infectious agents, foreign antigens or aberrant expressed endogenous antigens. Thus, understanding the molecular mechanisms of innate immune responses to exogenous and endogenous antigens would be able to develop novel strategy to treat numerous human diseases such as infection, autoimmunity, and allergic reactions. We developed innate immunity receptor array (IIR-EIA) and C-type lectin receptor array (CLRA) to identify receptors critical in virus-induced cytokine storm and host-defense mechanism. We are systemically identifying the functions of novel CLRs by gene-deficient mice, and generate human antibody against CLRs and other IIRs potential therapeutic agents to treat human diseases. We have identified the spleen-tyrosine kinase (Syk)-coupled C-type lectin receptors CLEC5A and CLEC2 are critical receptor in dengue virus-induced cytokine storm and thrombocytopenia, and antagonistic mAbs against CLEC5A and CLEC2 are being developed for therapeutic agents.

HsiehSL Host Pathogen Interaction

Immunomodulation

We develop serial recombinant proteins to skew monocyte differentiation into M2 macrophages to promote tissue regeneration and repairing, as well as attenuate septic and aseptic inflammatory reactions. Two recombinant proteins (DcR3.Fc and HBD.Fc) have been shown to induce M2macrophage differentiation via epigenetic regulation. These proteins have potent immunosuppressive effect to attenuate beta-amyloid induced-neuroinflammation, promote neuronal regeneration, and preserve memory in an Alzheimer mouse model.
