RESEARCH INTERESTS:

Crystallographic Studies of Biological Macromolecules

Research in the Hsiao lab is to provide the detailed structural information, through the systematic crystallographic investigations that is essential for the understanding the structure-function relationships of the biological systems. Current interests include: 1) Structural and functional study of the outer and inner membrane proteins involved in the intracellular trafficking of newly synthesized polypeptides from chloroplasts. Recently, we have determined the 3-D structure of pea Toc34 in complex with GDP and Mg2+ at 2.0Å resolution. From the structure, we proposed a working model for the import of proteins into chloroplasts. In this model, GTP hydrolysis at the G-domains of Toc159 and Toc34 results in translocation of the preprotein across the envelope. 2) Structural and functional study of rimosomal replication proteins required for primosome assembly on the lagging strand to pry apart DNA duplex and synthesize RNA primers. 3) Crystallographic study of protein-glycolipid (Heparin & Sulfatide) interaction to gain insight into the protein-membrane recognition and interaction.

我們的研究工作主要是利用x-光晶體繞射方法來了解並研究蛋白質的結構與功能機制的關係.目前的研究方向為:1) 葉綠體外膜及內膜轉運機組蛋白的結構與功能研究. 2) 引物合成體(primosome)中所含參與DNA複製所需蛋白質(PriA, PriB, PriC, DnaB, DnaC, DnaT 及 DnaG)的結構與功能研究. 3) 蛋白質與醣基化磷脂球(例如醣胺素等) 之交互作用與功能研究
